Familial Risk in Cancers of Unknown Primary

What are the risks for first- and second-degree relatives?

Relatives of patients with cancers of unknown primary (CUP) are at increased risk of CUP and several other malignant neoplasms, including lung, pancreatic, and colon cancer, according to a study of 4,160 patients who received a diagnosis of CUP. Researchers found:

• First-degree relatives of patients with CUP were at an increased risk of CUP themselves (HR=1.35), as well as lung (HR=1.37), pancreatic (HR=1.28), myeloma (HR=1.28), and non-Hodgkin lymphoma (HR=1.16) cancers vs controls without CUP.

• With analysis restricted to relatives of cancer-free controls, additional risks for colon (HR=1.19), and bladder (HR=1.18) were seen.

• Second-degree relatives of patients with CUP were at slight increased risk of lung (HR=1.14), pancreatic (HR=1.17), breast (HR=1.09), melanoma (HR=1.09), and ovarian (HR=1.19) cancers.
